      补体在心肌缺血再灌注损伤中的研究进展

      Research Progress of Complement in Myocardial Ischemia-reperfusion Injury

      摘要:
      心血管疾病是人类疾病致死的首要病因,而在各类心血管疾病中,缺血性心脏病最为常见.对缺血性心脏病进行的常见治疗性干预是心肌缺血再灌注,其诱导的无菌炎症反应在心肌损伤和重构中起到重要调节作用.补体作为天然免疫系统的重要组成部分,参与抗感染与免疫调节.除了众所周知的氧自由基和白细胞的作用外,补体系统的激活也在再灌注后的炎症反应中扮演重要角色.实验研究表明,抑制补体通路或许能够挽救心肌组织的进一步损伤,补体系统包括一系列蛋白质,这也提供了许多在急性心肌梗死中药物介入的位点.系统性了解补体系统的生物学特征、在心肌缺血再灌注损伤中的作用机制研究和补体靶向治疗研究具有重要意义.
      Abstract:
      Cardiovascular disease is the leading cause of death among various human diseases ,among which ischemic heart disease is the most common.The common therapeutic intervention of ischemic heart disease is myocardial ischemia /reperfusion,which can induce ster-ile inflammation,thus playing an important regulating role in myocardial injury and remodeling .As an important part of a natural immune sys-tem,complement is involved in resistance to infection and immunity .In addition to the well-known role of oxygen free radicals and white blood cells,activation of the complement system is also one of the major contributors of the inflammatory reaction upon reperfusion .Experimental ev-idence suggests that tissue salvage may be achieved by inhibition of the complement pathway .As the complement is composed of a cascade of proteins,it provides numerous sites for pharmacological interventions during acute myocardial infarction .Systemic understanding of the com-plement system ,including the biology characteristics ,role in myocardial ischemia-reperfusion injury mechanism and molecule targeted therapy research is of great significance .
