罗格列酮对大鼠血管平滑肌细胞凋亡和转化生长因子分泌的影响

Effect of Rosiglitazone on secretion of transforming growth factor-β1 and apoptosis of vascular smooth musle cell of rat in vitro culture

目的 观察罗格列酮对大鼠胸主动脉平滑肌细胞(SMC)凋亡和转化生长因子-β1(TGF-β1)水平的影响.方法 采用罗格列酮处理体外培养的大鼠胸主动脉平滑肌细胞(SMC),用ELISA法检测培养基中转化生长因子-β1(TGF-β1)的水平,同时用流式细胞学和TUNEL法检测细胞凋亡.结果 罗格列酮诱导SMC快速分泌TGF-β1,在0.5 h内达到高峰,并且这种快速分泌可被GW9662逆转;罗格列酮诱导SMC凋亡存在浓度依赖,随着罗格列酮浓度的增加SMC凋亡率越来越高,100 μmol/L时SMC凋亡率达到最高,而超过100 μmol/L时,凋亡率并没有随着罗格列酮浓度的增加而增加,甚至有所下降,并且GW9662和anti-TGF-β1抗体均可逆转罗格列酮对SMC凋亡的诱导.结论 罗格列酮通过诱导的激活及转化生长因子β1(PPAR-γ)的快快速分泌促进血管平滑肌细胞凋亡.
